Prepare a 1.5% agarose, 0.5x TBE buffer gel in a glass flask. Calculate how many agarose grams you will need to make a total volume of 150 mL. COver the top of the glass flask prior to heating. This is usually done with paper towels. Microwave the flask at high power for about 2 minutes or until all the agarose is dissolved. Remove the flask from the oven. Allow the gel to cool to 40 celsius, or when it is comfortable to touch, and then add an intercalating agent, such as 2 microlitres of ethidium bromide. Mix and pour into the prepared gel tray.
